Quality and compliance

Secure your GMP compliance. Control your contamination risks. Prepare for your inspections.

Axys-Network supports you in assessing, structuring and improving your contamination control strategy (CCS), in line with the requirements of GMP Annex 1, through three priority areas:

  • Regulatory compliance: identify and address gaps against Annex 1 requirements;
  • Risk control: a CCS that is consistent, documented, scientifically justified and operational;
  • Industrial performance: optimise aseptic processes, flows, practices and environmental monitoring systems.

Data integrity and computerised systems

What the agencies expect

Agency observations on computerised system validation (CSV) are rising: they now account for a quarter of all observations issued. Neglecting these systems exposes you to severe findings, particularly for major systems such as ERP, laboratory equipment and LIMS. Cyber risks are also receiving growing attention.

Binding standards: GMP, Annex 11, 21 CFR Part 11, ALCOA+ principles. Guidance: GAMP 5, ICH Q9.

Our approach

  • audit based on your documents (validation master plan, qualification files) and on actual practice on the shop floor, against the standards;
  • assessment of your risk analyses and of the depth of testing they require;
  • consideration of cyber risks;
  • prioritised action plan following the gap assessment, and follow-up of its implementation.

The most frequent gaps

What we have encountered most often over the past five years:

  • shortcomings against ALCOA+ principles: poorly locked analytical methods, backup and restore that fail or have never been tested, no second-person verification in hybrid set-ups (paper and computerised system);
  • complacent risk analyses that lead to edge cases not being tested;
  • cyber risks that are poorly assessed and insufficiently covered.

Visual inspection

Our experts support you on the visual inspection of your injectable products:

  • defect selection and criticality classification;
  • visual inspection qualification for manual, semi-automatic and automatic inspection;
  • visual inspection qualification for clinical batches;
  • difficult-to-inspect parenterals strategy;
  • statistical control and trend analysis.

Expertise · France

Site remediation after a difficult ANSM inspection

Context: following a difficult ANSM inspection, a French laboratory had to bring its site back into compliance.

Intervention: 4 Axys-Network experts deployed for 3 months alongside the site teams.

  • review of the notes from the inspection close-out meeting and assessment of the criticality of the observations;
  • set-up of the action plan and review of the draft responses to the ANSM;
  • roll-out of the responses on site before the final assessment;
  • participation in conference calls with the ANSM as technical subject-matter expert.

Result: the commitments made to the ANSM were in place on site before the final assessment. A global remediation plan was then drawn up to prepare the group's other sites.

Expertise · Japan

Auditing a Blow-Fill-Seal site before export to Europe

Context: a Japanese laboratory wanted to export a Blow-Fill-Seal product to the European market and reduce its dependence on European contract manufacturers.

Intervention: audit of the manufacturing site against European requirements, and recommendations to secure the project.

Result: the laboratory now exports its product to Europe.
